Objectives:( 1 ) To observe whether rat bone marrow harbors islet-like cells in hyperglycemic state.( 2 ) To establish the isolation and culture method of mesenchymal stem cells(BM-MSCs) and confirm their biological features.( 3 ) To induce rat BM-MSCs to differentiate into islet-like cells in vitro and evaluate their biological functions.( 4 ) To study the capacity of BM-MSCs differentiating into insulin- positive cells in diabetic rat in vivo.( 5 ) To investigate the protective effect of BM-MSCs on the function of islet grafts. Methods:( 1 ) Prolonged and transient hyperglycemic state was setup in SD rats induced by i.p. streptozotocin(STZ) or 50% glucose solution. Age-matched normal SD rats were used as controls. Rat bone marrow cells were isolated and cultured in low glucose DMEM supplemented with 10% fetal bovine serum (FBS). Cell morphology was observed under converted microscopy. Intracellular protein expression of islet-related hormones was observed by immunofluorescence under laser con-focal microscopy.
