1. 11, 12-epoxyeicosatrienoic acid Preconditioning in the Isolated Immature Rabbit HeartObjective: To study the effect of 11,12-epoxyeicosatrienoic acid (11,12-EET)preconditioning in protecting myocardium of immature hearts from ischemic reperfusion injury.Methods: On the Langendorff perfusion apparatus, isolated perfused immaturerabbit hearts underwent 120 minutes of normothermic ischemia, followed by 60 minutes of reperfusion. 24 isolated immature rabbit hearts were divided into three groups randomly: control group, ischemic preconditioning group (IPC) and 11,12-EET preconditioning group (EET). The preischemia and postischemia myocardial function were assessed by the percentage recovery of left ventricle developed pressure (LVDP), left ventricle end-diastolic pressure (LVEDP), ±dp/ptmax, myocardial water content (MWC), and coronary blood effluent (CBE). The myocardial ultrastructure and myocardial structure of light microscope, the index of arrhythmia scores (AS), creatine kinase (CK) and lactate dehydrogenase (LDH) were also observed.Results: (1) The percentage recovery of myocardial function of IPC and EETgroup were significantly better than control group. No structure of light microscope and ultrastructural changes were fund in the EET group and IPC group while myocardial structure injury were observed in the control group obviously. (2) The value of CK,LDH and myocardial water content of EET group were lowed than that of IPC group, at the same time, coronary blood effluent of EET group was higher significantly.Conclusion: Ischemic and 11,12-EET preconditioning can provide more better myocardial protection for ischemic reperfusion immature rabbit hearts.
