Objective: Interstitial pulmonary fibrosis is a series of diseases characterized by early alveolus lesion, inflammatory cells infiltration, followed with collagen aggradation and finally changed into interstitial fibrosis. They have the same clinical features such as cough, progressive dyspnea, a typical honeycomb aspect on the high-resolution CT-scan, restrictive ventilation disturbance, decreace of diffusive function, hypoxemia and respiratory failure. Most patients die 3 to 8 years after diagnosis. Hunting effective therapeutic method such as traditional Chinese medicine is a hot problem because of increasing incidence, unknown mechanism and lack of effective method in west medicine. In this article, Huaxian decoction (HD), a prescription of tonifying qi and regulating yang, smoothing qi activing blood and eliminating sputum, was chosen to treat lung fibrosis rat model and its effect and mechanisms were observed and investigated.Methods: One hundred and forty-four healthy Wistar rats weight from 180 to 240gram were randomly divided into six groups(n=24, each): normal control group (group A), model group (group B), prednisone group (group C), Chinese drugs of high, moderate and low dosage (group D, E, F). The rat model was induced by blyomicin and respective medicine was given to each group. Every group was subdivided into three groups (n=8) which was killed in the 7th, 14th and 28th day respectively. ①The rat's lung tissue section was observed under light microscope and electron microscope to study the morphological alternation. ②ELISA method was used to assay the sera level of TGFβ1 and IL-4. ③TBA method and colorimetric method were used to detect the content of MDA and GSH in lung tissue respectively. ④The expression of TGFβ1 in lung tissue was measured by the method of RT-PCR. ⑤The lung tissue was made into suspending liquid and colored with Rh123 and PI respectively. The fluidity of mitochondrion membrane was detected with flow cytometry. Results: ① Changes of pathologic morphosis under light microscope in the rat lung fibrosis model: in the 7th day after model was made, interval between alveolus became thicker, , the number of fibroblast increased and alveoli in the focus of infection were atrophied and lots of inflammation cells infiltrated; in the 14th day after model was made, interval between alveolus became thicker and thicker, fibroblast and collagen matrix increased markedly; in the 28th day after model was made, alveoli structure was damaged, the wall of alveoli became thicker remarkably, interstitial fibrotic cicatrix came into being and capillary blocked. Changes under electron microscope in the 7th and 14th day showed that mitochondria of type Ⅱ alveolar (epithelial) cells became hyperplastic and swollen and its cristae decurtated, collapsed and vanished.
