It has been proved that Actinobacillus actinomycetemcomitans(A,a), especially it's surface-associated matecials (SAMs) plays an important role in juvenile periodontitis(JP). However the mechanism of the pathogenesis of JP remains unknown.Part Ⅰ1. Extraction and purification of SAMs from A.a. SAMs of A.a were extracted with saline and purified by HPLC. Preliminary chemical analysis of the purified SAMs indicated that it is mainly glycoprotein and little endotoxin. SDS-PAGE analysis revealed that there were several bands on PAGE profiles, suggesting the components of SAMs were quite complicated.2. Examination of cytotoxic activity of SAMs against fibroblast L929. Cytotoxic activity of SAMs against fibroblast L929 was examined with MTT assay, cell counting and image analysis. The results showed that SAMs exhibited a pronounced increase in cytoplasmic and nuclear area, inhibited division and proliferation of L929 cells in a dose-dependant manner, at the concentration of 100μg/ml the inhibitary effects was significant compared with the control(P<0.05).
