Objectives: (1) To investigate the effects of raloxifene on the growth of Human breast cancer cell lines MCF-7, MDA-MB-231, human normal breast epithelial cell line HBLIOO and human ovarian cancer cell line SKOV3; (2) To explore the mechanism of cell growth affected by raloxifene.Methods:Part one: Growth characteristics of human breast cancer cell lines MCF-7, MDA-MB-231 and HBL100 were described with morphological observation and cell growth curve.Part two: The effects of raloxifene on the growth patterns, cell cycle of MCF-7, MDA-MB-231 were studied with MTT assay and FCM respectively. Comparing with tamoxifen, 17β- E2 and progesterone, we study the mechanism of these effects. Furthermore, The expression of Caspase 3 in raloxifene- treated MDA- MB- 231 was analyzed by ICC and the effect of progesterone on VEGF mRNA level in MDA-MB-231 was studied by ISH. The effects of raloxifene on the growth of HBL100 was studied by MTT assay and inverted microscopy.Part three: Compared with tamoxifen, 17β- E2 and progesterone, the effects of raloxifene on human ovarian cancer cell line SKOV3 were studied by MTT assay. The expression of Ki-67 in SKOV3 cells administrated with different chemicals were evaluated by ICC.Part four: The expression of ER, PR in raloxifene- treated SKOV3 cell was studied by ICC. The ability of raloxifene, tamoxifen, 17β- E2 and progesterone to increase the expression of ER mRNA and protein in SKOV3 cell with c- ELISA, RT-PCR and western blotting was examined.Results:Part one The growth of MCF-7 and HBL100 is close related to the...
