Objective:1. To use streptozotocin to produce the experimental diabetic rat model which has high expression of RAGE-mRNA in cardiac muscle and aorta.2. To use the method of in situ hybridization to observe the effect of Xianzhen Tablet on high expression of RAGE mRNA of cardiac muscle and aorta in diabetic rat, using aminoguanidine as a control drug and supply experiment basic for clinic application of Xianzhen Tablet.Materials:1. Animal: Male Wistar rat with 7 weeks old, weighing 180-220g.2. Main drugs and agents: Xianzhen Tablet, aminoguanidine, streptozotocin, and non-radial DNA-labeled kit.3. Main instruments: Blood glucose instrument of One Touch II, RT-PCR instrument, instrument of RNA retraction, Olympus microscope, and computer micro-imaging system.Methods:1. Production of diabetic rat model: to inject streptozotocin (60mg/kg) into left and low part of peritoneal cavity at one time to induce the formation of the diabetic rat model ( Blood glucose is above 16.7mmol/L)2. Grouping and administration methods: All rats were randomly divided into three groups: treated diabetic group with Xianzhen Tablet (herb 20g/kg/d) treated diabetic group with aminoguanidine (100mg/kg/d) and untreated diabetic group (water).
