ObjectiveOn the basis of traditional Chinese medicine theory, the damp-heat syndrome rat model was established by compound causes. And the model was evaluated by observing the changes of immunity, metabolism function, and disproving with corresponding Chinese medicines. On the basis of above model, the pathopoiesia mechanism of damp-heat syndrome model and therapy mechanism of Qingre Zaoshi Prescription were explored by observing the express of the cell endotoxin (ET) specificity receptors, such as LBPmRNA, CD14mRNA, TLR4mRNA, NF-κ Bp65, etc.MethodsIn respect of model establishment, the damp-heat syndrome model was eatablished by compound factors, the treated by Qingre Zaoshi Prescription. The secretion type immune globulin A(SIgA), interleuin-1 (IL-1), tumor necrosis factor-α (TNF-α ), motillin(MTL) and gastrin(GAS) were detected by radio-immunity measures; the high density lipoprotein-cholesterol (HDL-c), low density lipoprotein-cholesterol (HDL-c) were detected directly, and the active of liver mitochondria Na~+-K~+-ATPase was also detected to evaluate the model. Then using the above model, the expression changes of LBPmRNA, CD14mRNA and TLR4mRNA were semiquantitated by reverse transcription -polymerase chain reaction ( RT-PCR) , the active of nuclear factor-kappa Bp65 (NF-κ Bp65) were detected by immune-histochemical methods in order to make clear of the action spots of endotoxin transduction signals interrelated protein molecules receptors in the interference of Qingre Zaoshi Prescription on damp-heat syndrome rats, then to explore the pathogenesis of damp-heat evil from Cellular and Molecular Biology levels.Results1 the establishment of damp-heat syndrome modelThe results show that the changes of symptoms and signs in model group rats were correspond with traditional Chinese medical science damp-heat syndrome. The contents of SIgA, TNF-α and IL-1β in model group were remarkably increased, but has no significant difference from the contrary group; the contents of HDL-c, LDL-c in model group were remarkably increased, and HDL-c has significant difference from those in contrary group; the contents of MTL were remarkably increased and the contents of GAS were remarkably reduced in model group, and has no significant difference between those in contrary group, and the contrary group and the normal group has no significant difference: the Na~+-K~+-ATPase active...
