Part I The establishment of an inflammatory breast caner cell line expressing dominant negative E-cadherin Mutant(H-2kd-E-cadherin)Objective: To establish an inflammatory breast caner cell line model which expressed dominant negative E-cadherin Mutant(H-2kd-E-cadherin).Method: Dominant negative E-cadherin Mutant cDNA was introduced into an Human inflammatory breast caner cell line (SUM 149) which expressed higher levels E-cadherin. Several G418-resistant clones were obtained.Results: Using RT-PCR and FITC-conjugated Flow cytometry analysis methods, we identified two G418-resistant clones which expressed higher levels dominant negative E-cadherin Mutant. Further studies showed the endogenous E-cadherin of the two clones was significantly down-regulated, as compared to the controls (the parental SUM 149 cells and Mock-transfected SUM 149 cells)Conclusion: The SUM 149 cells which expressed dominant negative E-cadherin Mutant provide us a suitable cell model to study the E-cadherin function on the Human inflammatory breast caner cell line SUM149.
