| Ageing Is the basis of geratobgy medk,ne. Although the process of ageing as not pathologic, it has the potential to develop many kinds of diseases related to age. Many people have made efforts for such research, however the mechanism of ageing is not quite clear so far, and therefore the treatments are also not effective, safe and convenient as expected. The injury to the body of ageing is progressive, usually irreversible, so the ideal method of prevention in the early stage of ageing is the highlight of geratology. My mentor, Prof. Li Xuewu has abundant clinical experiences, especially with the moxibustion for health care. Tianjiu features as convenience~ cheapness and better effects, which has a developing perspective of clinical application in the future. Based on the situation of no study on the antagonism of lianJiu to ageing, a trial is carried out with lian.Jiu Plaster. The theoretical foundation as ROS (Reactive Oxygen Species) - apoptosts -ageing, and the animal models of acute ageing are made with D-gal, whose startup factor is oxygen free radicals. Our trial focuses on the effects of innovated Tianjiu Plaster with the controls of traditional moxibustion, and the results relate to the biochemical, morphological, cellular and molecular changes of neurons, which suggest the influences of lianJiu on ageing injuries. Intelligence test In the learning test with water maze and platform, the models, in contrasts with the controls, show an increase of learning impairment that the time of completing task and the times of errors are significantly prolonged. The models treated with lianJiu Plaster show an significant increase of learning in contrast with the controls, which indicates the better effect of TianJiu than the traditional moxibustion on the dysfunction of brain induced by ageing. 2. Biochemical results: in the brains and livers of the models, the oxygen free radicals and intracellular cabum influx increase, and the degree of ATh decreases. All these demonstrate the oxkiative stress is the startup of aging. In the models treated with TianJiu Plaster, the activity of antioxidase SOD is increased, the degree of MDA partly decreased, and the excessive intracellular calcium reduces. These results indicate the plasticity of cellular metabolism and antagonism to the course of aging. 3. Ulfrastiuct ral changes: the number of neuron decreases in the CAl of hippocampus in models and the neurodegeneration involves the degeneration of nuclei, cellular tumidness and vacuolation of mitochondnon. TUNEL shows the apoptosis that occurred in the loss of neurons. The TianJiu PIaste~s improving the conditions above indicates that TianJiu can antagonize the neuronal degeneration caused by D-gal, and help the recovery of neurons. 4. Thechangesof apoptosis The expression of bd-2 was not found in the apoptotic genes, and the least in the neurons of models, which reveals the dues of the relationship between the apoptosls of ageing neurons and the regulation of the expression of gene. T~nJiu is able to antagonize apoptosis by mediating the expression of anti-apoptosis gene. The result of our study suggests that the models induced by D-gal have the neurobicchemcal, subceliularly pathological, neurodegenerative and molecularly biological features. And the mechanism is that the increasing free radicals can cause the overfreight of calcium in the neurons, the dysfunction of energy metabolism and other damages, which lead to the neuronal agei... |
