| Objective:With the observation of the effects of nephritis Xiaobaikeli towards the urinary protein in24hours, kidney, cell factors, prodocyte-relating proteins and p38MAPK Signal Transduction Pathway of the rats of diabetic nephropathy (DN) at the early stage in experimental research, its mechanism in delaying the development of DN is investigated.Method:After Unilateral nephrectomizing the models of rats of diabetic nephropathy at the early stage with High Fat/High Sucrose Diet and Streptozotocin, they are separated into5groups:the blank control group, the model contrast group, the group with high dosage of nephritis Xiaobaikeli, the group with low dosage of nephritis Xiaobaikeli and the group with Benazepril Hydrochloride (Lotensin). After8weeks, test the weight of the rats, their weight of kidney blood sugar, blood serum creatinine (Scr),blood urea nitrogen (BUN) and UAE; test the content of the advanced glycation endproducts (AGEs) with ELISA; Color the Pathological Sections PAS, observe the Pathological changes of the kidney of the rats; Look into the expression of cell factors TGF-β1and CTGF with the Immunohistoche mistry; test the expression of a-actinin-4mRNA, podocalyxin-mRNA and p38MAPK-mRNA of the rats'kidney through Semi-quantitative RT-PCR; test the expression level of α-actinin-4,podocalyxin and p-p38MAPK with Western Blot.Result:Nephritis Xiaobaikeli fails to ameliorate the weight of the rats of diabetic nephropathy at the early stage, their weight of kidney blood sugar, blood serum creatinine (Scr) and blood urea nitrogen (BUN),with no distinctive difference compared with the model group (P>0.05); Nephritis Xiaobaikeli can reduce the weight of the kidney and decrease its index; it can reduce the urine UAE evidently, the group with nephritis Xiaobaikeli is much clearer than the model group (P<0.05), it can lighten the Mesangial Proliferation, Basement Membrane incrassation and the fuse of foot process; it can decrease the expression of TGF-β1,CTGF of the rats'kidney, with distinctive difference compared with the contrast group (P<0.05); it can increase the expression of α-actinin-4mRNAand podocalyxin-mRNA of the kidney, as well as decreasing the expression of p-p38MAPK-mRNA, both with distinctive difference compared with the contrast group (P<0.05); it can increase the expression level of α-actinin-4,podocalyxin and p-p38MAPK with Western Blot, both with distinctive difference compared with the contrast group (P<0.05)Conclusion:Nephritis Xiaobaikeli can relieve the kidney index of the rats of diabetic nephropathy at the early stage, reduce the content of the AGEs, with the increasing of the expression of prodocyte-relating proteins a-actinin-4and podocalyxin, the decreasing of the level expression of the rats'kidney p-p38MAPK, the restrain of p38MAPK Signal Transduction Pathway, the reducing of the expression of rats'kidney cell factors TGF-β1and CTGF and the reducing of the discharging of the Proteinuria, the purpose of protecting kidney is abstained, at the same time, the diabetic nephropathy of the rats is delayed. |
