Cancer is a class of diseases with uncontrolled growth, invasion andmetastasis characters, causes about 13% of all human deaths worldwideevery year. Since cancer metastasis is the major cause of cancer mortality,a great deal of attention has been led to discover potential pharmaceuticalcompounds as anti-metastasis candidates. The marine environment is fullof natural compounds with amazing biological and pharmacologicalactivities. About 80% of the planet's animal and plant reside in the marineenvironment. Up to present, more than 20,000 new compounds have beenisolated from marine organisms. The marine-derived algae, one importantmember of marine organisms, have been regarded as a fresh source ofnatural products with significant therapeutic potential.In the present study, Dieckol (118.78 mg) and 6,6′-bieckol (172.51mg)were isolated from marine-derived algae Ecklonia Cava in Korea in 2010.These two compounds did not show any significant cytotoxic effect in human fibrosarcoma cells (HT1080) 250μM. However, compoundsinfluenced on migration and invasion of HT1080 cells in aconcentration-dependent manner, and the number and length of stellatestructures of HT1080 cells were significantly reduced in 3D culturecondition by the treatment of compounds. Moreover, the expressions ofMMP-2,-9 were inhibited in a dose-dependent manner in HT1080 cells.AP-1 and NF-κB are major transcription factors that regulate MMP-2 and-9 expressions. Dieckol and 6,6′-bieckol significantly inhibited p65 andp50 nuclear translocation, but not ERK,p38 and JNK phosphorylationand c-jun transactivation.Dieckol and 6,6′-bieckol have inhibitory effect on migration andinvasion of cancer cells in present study,it is suggested that thesecompounds have potential to be utilized in nutraceutical and therapeuticagent for anti-metastasis therapy.
