Part□Pentamethylquercetin improves adiponectin expression in 3T3-L1 adipocytesAim:To investigate the effects of PMQ on the adiponectin mRNA and protein expressions in 3T3-L1 adipocytes. Methods:3T3-L1 preadipocytes were differentiated to mature adipocytes. The adipocytes were incubated with PMQ (0.1-10μM) for 24 hours ans 3μM PMQ for 0,3,6,12,24,48 hours. RT-PCR and western blot used to found the semi-quantatative values of adiponectin. Results:□;□PMQ increased adiponectin mRNA (P<0.05) expression at the concentrations of 1,3 and 10μM, but not at 0.1 and 0.3μM. Results from western blot analysis also confirmed the dose-dependent change in adiponectin protein expression after PMQ treatment PMQ increased adiponectin mRNA and protein expressions in a time-dependent manner. Conclusion:The herein presented data demonstrated that a synthetic polymethoxylated flavonoid pentamethylquercetin (PMQ) improved adiponectin mRNA expression and protein level in 3T3-L1 adipocytes. This study raises a novel finding that PMQ could be a promising new candidate for the treatment of metabolic diseases in the future.Part□The potential mechanisms of PMQ improves adiponectin expressionAim:To investigate the mechanisms of PMQ improves adiponectin expression in 3T3-L1 adipocytes. Methods:Mature 3T3-L1 adipocytes were incubated with various concentrations of PMQ for 24 hours. RT-PCR and western blot used to found the semi-quantatative values of PPARy, phosphorylated Akt and total AKT, TNF-a, IL-6. At day 10. Adipocytes were incubated for 24 hours with PMQ (3μM), GW9662 (10μM), wortmannin (100 nM), PMQ+GW9662, PMQ+wortmannin respectively, RT-PCR and western blot used to found the mRNA and protein expressions of adiponectin. Resuls:□PMQ upregulated PPARy mRNA and protein expressions in a dose-dependent manner.□PMQ significantly increased phophorylated Akt level without affecting total Akt.□Wortmannin or GW9662 itself has no effects on the adiponectin expression. However, Wortmannin and GW9662 can significantly antagonize PMQ-induced increase of adiponectin expression in mRNA and protein levels.□PMQ decreased expression and secretion of TNF-a and IL-6 in a concentration-dependent manner. Methods:3T3-L1 preadipocytes were incubated with different concentrations of 5-PBDMA during the differentiation process, at day 10, RT-PCR used to found the semi-quantatative values of adiponectin, PPARs, TNF-a, IL-6, FGF-21. Results:□5-PBDMA has no effect on PPARs mRNA expression;□5-PBDMA increased adiponectin mRNA (P<0.05) expression at the concentrations of 0.1,0.3μM, but 5-PBDMA 1,3μM decreased the mRNA expression of adiponectin;□5-PBDMA decreased mRNA expression of TNF-a and IL-6 in a concentration-dependent manner in 3T3-L1 adipocytes; 5-PBDMA has no effect on FGF-21 mRNA expression. Conclusion: The herein presented data demonstrated that 5-PBDMA enhanced adiponectin mRNA expression and decreased TNF-a and IL-6 mRNA expressions in 3T3-L1 adipocytes.
