| Weining capsule is used to treat epigastric pain, enjoying massage and vomiting acidic liquid induced by disharmony of liver and stomach, Qi deficiency of spleen and stomach; to treat gastric ulcer and stomach pain that have the above symptoms. In our studies, the pharmacodynamic action of Weining capsule was studied from angles of anti-ulcer, antispasmodic analgesic, antiemetic, anti-inflammatory and hemostatic. We discussed the mechanism of its anti-ulcer. Our tests provide some reliable pharmacological proofs for the clinical application of Weining capsule in treating gastric ulcer and stomach pain. The results are showed as followings:1. The therapeutic action of Weining capsule on gastric ulcer:Weining capsule showed significant anti-ulcer effect on rat pyloric ligation. Its treatment effect on pyloric ligation ulcer model in rats is multifaceted as following:Weining capsule showed the significant inhibition on gastric juice volume, total acidity of gastric juice and pepsin activity and reduction of the pathological changes of gastric ulcer. Weining capsule can reduce the ulcer index of indomethacin plus ethanol-induced gastric ulcer in mice, however, for reserpine gastric ulcer in mice, can reduce ulcer bleeding points.2. The effect of antispasmodic analgesic:Weining capsule showed significant inhibiting effect on gastric emptying. Moreover, Weining capsule can significantly increase the weight percentage of residue in stomach. Three different dosages of Weining capsule showed significant inhibiting effect on promotion of small intestine motor function in mice. Weining capsule significantly inhibited the contraction frequency of ileum movements in rat hyperactivity induced by acetylcholine in the body. Weining capsule showed significant analgesic effects. Hot plate analgesia test results showed that Weining capsule significantly increased the pain threshold in mice. Writhing test results showed that Weining capsule can prolong the incubation period of writhing occurs and reduce the writhing occurrence wihtin 20 minutes. Weining capsule showed significant sedation. It could significantly reduce the frequency of spontaneous activity in mice.3. The antiemetic effect:Weining Capsule showed significant antiemetic effect on copper sulfate induced vomiting in pigeons. It can significantly prolong the latent period of vomiting and reduced the frequency of vomiting.4. The effect of anti-inflammatory:Weining capsule has anti-inflammatory effects. It significantly inhibited xylene-induced mouse ear inflammation and the formation of granuloma in mice.5. Hemostasis:Three different dosages of Weining capsule can significantly shorten the bleeding time and clotting time of the mice.6. The expression levels of HSP-70, EGF and EGFR mRNA in Rat gastric mucosa, content of GAS in Serum and contend of PGE2 in gastric mucosa were regarded as the predominant indexes to study the anti-ulcer mechanism of Weining capsule. Our studies showed that the mechanism of Weining capsule on inhibiting the gastric ulcer lied in the following facts:it can increase the expression levels of HSP-70, EGF and EGFR mRNA of gastric mucosa. It can significantly reduce the levels of serum GAS and significantly increase PGE2 content in gastric mucosa in rats.
