Objective To identify the expression of microRNAs and predicted mRNA during adipogenesis, osteogenesis and endothelial differentiation of Adipose tissue - derived mesenchymal stem cells. Methods Expression of these selected microRNAs and their predicted gene were examined by RT-PCR, Real-time PCR. Some related protein was also examined by Western blot. Results Changes of the expression pattern of these microRNA and their predicted gene were analyzed according to the reported functions of predicted gene and their signaling pathways. miR-K and miR-M were definitely down regulated during adipogenesis, while they remained unchanged during osteogenesis and endothelial differentiation. Conclusion We suppose that miR-K and miR-M may play important roles in regulating adipogenesis. The predicted target gene of miR-K is Akt1, while that of miR-M is Notch1.
