Spermatogenesis and spermiogenesis are a series of highly ordered process of molecular and cellular events. The spatial and temporal regulation of gene expression is very complicated. More and more cDNAs encoding sperm-related proteins involved in these two processes have been identified and characterized.In previous studies, a rabbit sperm tail protein with an estimated MW of 20KD was purified and designated as rSMP-B. Monoclonal antibodies (mAbs) were raised against the rSMP-B protein and demonstrated to interact with human as well as rat sperm proteins with an estimated MW of 70KD. These mAbs inhibited the penetration and fertilization of Zona-free hamster eggs by human sperm. Adult male rabbits were immunized with rSMP-B, their spermatogenesis was blocked at spermatid stage. It suggested that rSMP-B might play an important role in spermatogenesis and spermiogenesis. A 2.0Kb cDNA (RSD-1) was obtained by screening a rat testis A.gt11 cDNA expression library with mAbs against rSMP-B. Its sequence was determined and submitted to GenBank with accession number U26723.In the present study, a RSD-1 2.0Kb cDNA was used as a probe to screen full length cDNA from human testis λgt10 5'-stretch and λ ZAPII cDNA libraries respectively. 12 positive clones were obtained and confirmed by Southern blot.
