Studies On The Cutaneous Pharmacokinetics And Metabolites Of The Alkaloids In Rats

Study on drug metabolism plays an important role in drug research and development. Recent years, metabolite analysis for chemical constituents of traditional Chinese medicine also attracts a great interest. We have carried out some research on three alkaloids: sinomenine, matrine and oxymatrine, detailed work as follows:The microdialysis auxiliaries (animal body temperature pad, linear probes, vascular probes etc.) were self-designed and made. The in vitro and in vivo performance of the microdialysis technique was established for the study of sinomenine, oxymatrine and matrine. The phamracokinetics and biotransformation of sinomenine in rat skin microdialysates and plasma were also investigated by LC-ESI-MS. A specific and sensitive liquid chromatography tandem mass spectrometry (LC/MS/MS) method was established for the determination and pharmacokinetic study of oxymatrine and its metabolite matrine in rat blood and skin microdialysates after the topical application of oxymatrine gel (3% oxymatrine).All of these studies provide scientific data and enlighten the thought for the modernization of Traditional Chinese Medicine.
