Objective: 1. Researched and Developed stereo-directional fixation system for smallanimal and spinal cord injured tube. 2. Established standard operationalprocedures. 3. Established an experimental fluid-percussion animal model ofsemi-spinal cord injury in the rat. The model should be success.fullyreproduces, stable and different grades of injury degree. 4. Evaluated spinalcord function and injury grade with several items (include BBB score,electric physiology, MR and pathological examine. This model will be thefoundation of further studies about complex physiologic and histo-pathologicchanges after SCI and proof-test new treatment methods associated with SCI.Methods: 1.64 SD rats were divided into two groups, the experimental group(n=48)andcontrol group (n=16). The experimental group was divided into 3 subgroups(n=16 for every subgroup). 2.Both two groups undertook standardoperational procedures; bone window was made at fight side of T10vetebra.3. Established three grade (mild, middle, severe) spinal cord injuryanimal model with fluid percussion injury system and stereo-directionalfixation system.4.Evaluated with neurological function score, motor evokedpotential,MRI,pathological and Immuno-histochemical methods at3days, 7days, 14days after injury.Results: 1. The animal could be stably fixed on stereo-directional fixation system and all joints of injury tube were sealed well, no water leaked during blowmoment.2.Neurological function score, motor evoked potential presentedSignificance differentia between every grade (P<0.05)along with theincreasing of injured energy. The area of injury enlarged with theincreasing of injured energy through MRI and pathologicalcheck.3.Incompletely neuron-function recovery could be seen along withtime prolonged.4. Less ratio of intercurrent syndroms occurred in this animalmodel.Conclusions: 1. A well effecting had been seen with using the stereo-directionalfixation system for small animal and spinal cord injury tube, elevatedveracity and reproduction.2.The present spinal cord injury animal model wasa clinic mimic, highly reproduced and was good at grade controlled. 3.Theinjury grade could be exactly evaluated with neurological function score,motor evoked potential,MRI,pathological and Immuno-histochemicalmethods.4. This model had less ratio of intercurrent syndromes and highlong-life ratio.
