Purpose:To explore the pathogensis of endometriosis and therapeutic mechanism of TKS.Method:At first,we created rats model of EM.On this basis we observe the ultrastructure and histomorphology by light microscope and electron microscope, investigate the. level of PGE2 and 6-keto-PGF1α in serum with RIA and the positive expression of E-Cadherin,MMP-2,VEGF and COX-2 protein in the focal tissue with immunohistochical method, determine the apoptosis of local tissues and cells by the approach of TUNEL in different rat groups.Result:1.The level of PGE2 in serum was higher evidently in EM group than that in normal group(P<0.01), while the level of 6-keto-PGF1α was lower evidently(P<0.01).2.The positive expressions of E-Cadherin in the focal tissue were lower in EM group than that in normal group evidently(P<0.01).3.The positive expressions of MMP-2 in the focal tissue were higher in EM group than that in normal group evidently(P<0.01).4. The positive expressions of VEGF in the focal tissue were higher in EM group than that in normal group evidently(P<0.01).5. The positive expressions of COX-2 in the focal tissue were higher in EM group than that in normal group evidently(P<0.01). the correlation between COX-2 and E-Cadherin in EM group was positily,while the correlation between COX-2 and MMP-2,VEGF in EM group were both negatively.6. The positive expression rates of PCD in the focal tissue were lower in EM group than that in normal group evidently(P<0.01).Conclusion:1. TKS can decrease the level of PGE2 and increase the level of 6-keto-PGF1α in serum EM group rats,therefore TKS can improve the blood circulation and reduce the synthesize of estrogen to inhibit the growth of EM.2.TKS can improve the positive expressions of E-Cadherin protein in EM rats' heterotopic endometrium,therefore TKS can improve the adhension ability of local tissue and reduces the incidence of EM invading and transferring.3. TKS can decrease the positive expressions of protein MMP-2 in EM rats' heterotopic endometrium,therefore they can not destroy basilar memgrane and decrease the ability of invading, transferring and implanting.4. TKS can decrease the positive expressions of VEGF protein in EM rats' heterotopic endometrium,so TKS can inhibit the formation of local vessel nets and decrease their blood supplying.5. TKS can decrease the positive expressions of COX-2 protein in EM rats' heterotopic endometrium, inhibit the formation and development of EM.6. TKS can improve the positive expressions of the apoptosis of local tissues and cells in EM rats' heterotopic endometrium,it can be deduced that TKS may take effect EM by inducing the apoptosis of heterotopic membrane cells.
