Abstract/Summary:
Quality control of Traditional Chinese Medicines (TCM) is bottlenecks in TCM modernization. In this dissertation, based on biothermodynamic theory, a novel method utilized in the research of screening bioactive substances and quality control of TCM was established.The inhibition effects of different extracts extracted from Folium isatidis (FII) and six anthraquinones on bacteria were investigated with biothermodynamics for the first time. The biothermodynamic results were consistent with the results of routine experiment. Therefore, the thermodynamic method was feasible for the expression of TCM bioactivity.FII could inhibit pathogenic microorganism and was in common use as alexipharmic and antifebrile drug. Firstly, the inhibitory effects of different extracts from FII on Escherchia coli and Staphylococcus aureus growth were studied by microcalorimetric method. The decoction and its water-extract of FII stimulated the growth of E. coli. While, the other four organic solvent extracts of decoction inhibited the growth of E. coli. The organic solvent extracts from decoction of FII slightly inhibited the growth of S. aureus.Additionally, the inhibitory effects of six anthraquinones (AQs) on S. aureus growth were quantitatively investigated by microcalorimetry. The experimental results indicated that the sequence of anti-microbial activities of the six AQs was: rhein>emodin>aloe-emodin >1,8-dihydroxyanthraquinone>chysophanol>physcion. All the results above-mentioned were matched with the conclusions reported in literatures. The biothermodynamic method was very useful by its extensive applicability and conveniency. It provided a new and practical method in the expression of bioactive pharmaceutical parts and substances of TCM.An integrated method for the extraction of organic acids, amino acids and polysaccharide from FII was investigated for the first time. Succinic acid, 2-amino benzoic acid, benzoic acid, salicylic acid and syringic acid in organic acid extracts of FII were determined with an HPLC method.To establish a novel method to evaluate quality of TCM, the idea of synthetic investigation of chemistry and pharmacology was brought forward in the dissetation. The correlation analysis of P-t curve of organic acid extracts and HPLC plots of five organic acids indicated the growth rate constants k1,k2 of bacteria inhibited by organicacid extracts were interrelated with contents of 2-amino benzoic acid, benzoic acid and salicylic acid of FII. Therefore, the method for evaluation quality of FII was set up by thermodynamics, and the biothermodynamic parameters of organic acid extracts of FII were as pharmacological evaluation criterion. Then, the quality evaluation method was utilized to estimate the FII samples and the results were consistent with other quality control methods. The conclusion suggested the thermodynamics could be utilized in evaluating TCM quality.To sum up, a novel method based on biothermodynamics was established and utilized in bioactivity screen and TCM quality control. The method combined with TCM theory and biothermodynamic theory provided a new point of view and a new methodology in the research of TCM modernization.
