| Allogeneic bone grafting is an alternative to autogeneic bone grafting currently in use, nevertheless, the immune rejection make allogeneic bone inferior to autogeneic bone in osteogenesis. How to eliminate the antigenicity and activate the osteoinductive ability of the the allograft is still an important problom in orthopaedic surgery. Experiments on decalcification and activation of osteoinductive ability, comparison between the osteogenesis of demineralized bone matrix in orthotopic and in heterotopic sites, and correlation between immune response to allograft of bone and osteoinductive activity of the graft, were reported in this paper. 1. Syngeneic demineralized bone matrix and the mixture of DBM with bone ash or acid-soluble protein extracted from demineralization was implanted into muscle pouch of mice, subsequently, the inhibition of DBM osteoinduction by bone ash or acid-soluble protein was demonstrated with histological and by means of alkaline Phosphatase assay. 2. Allogeneic demineralized bone matrix was implanted subperiosteally and subcutaneously in rabbits, six weeks postoperatively, more new bone formation in subperiosteal implants than subcutaneous ones was shown by histomorphometric quantification. 3.
