Dencichine, named β-N-oxalyl-L-a, β-diaminopropionic acid (β-L- ODAP) is the major non-protein amino acid which isolated from the seeds of Lathyrus sativus and detected in several other plants including other genera of leguminous and non-legume plants, Panax ginseng, Panax notoginseng and Panax quinquefolius. Dencichine has been proved to have strong homeostatic effect and can be used for the prevention and treatment of bloodloss in many hemorrhage models. Besides, it can increase the number of platelets and improve the symptoms of thrombocyto-pathy, thromabocytasthenia and hemorrhagic purpura caused by thrombocytopenia. Since dencichine displayes homeostatic effect in the lower dosage and toxic dosage is much higher than curative dosage, it is promising to exploit dencichine as a kind of high performance haemostat.The synthesized dencichine was the target compound in this manuscript. The structure of synthesized product was analyzed using a series of spectral techniques including UV, IR, MS, NMR and elemental analysis. Especially, the single-crystal X-ray diffraction and rotation determination which can provide valuable information about spacial structure were used for the identification of synthesized product with the natural extract. Furthermore, the systematic study on the controlling of synthesized dencichine quality was carried out. Due to the special nature of Den which has the classic amphoteric structure of amino acid, the strong polarity and no characteristic absorption in UV, for the assay test of Den, different methods were investigated respectively including acid-base titration, formal titration, nonaqueous titration, nitrogen determination and ion-pair HPLC. Among these methods, nonaqueous titration, nitrogen determination and HPLC could be used to assay dencichine successfully. The results were accurate and the precision was good.
