Background : Neo-adiuvant chemotherapy (NAC) which as the standard therapy project has been accepted widely. The survival ratio of patients with pathology pCR could be improved after Neo-adiuvant chemotherapy. Therefore, how to improve the NAC therapy effect and forecast the therapy effect were study emphases of Locally advanced breast cancer (LABC). In addition, RNA interference (RNAi) as new gene knock-out technique, could silence target gene at transcription after level and displayed enormous foreground in gene function and therapy studies.Objective : To improve the therapy effect of Neo-adiuvant chemotherapy, the EPI-ADM and PTX in two-weeks chemotherapy were used to therapy LABC, and the curative effect, toxicology effect and side effect of NAC, the pathology change of breast tumor were investigated. To seek the efficiency index of forecasting Neo-adiuvant chemotherapy, to affirm the forecast effect of hTERT to the therapy effect of Neo-adiuvant chemotherapy, to compare the relativity of the hTERT expression with the therapy effect of NAC before chemotherapy, and to compare the relativity of the hTERT expression with the therapy effect of NAC after chemotherapy, and afford the new index to forecast the therapy effect of NAC before chemotherapy.
