Research background and object:Stroke is a kind of polygenic disease caused by genetic and environmental factors.Epidemiological survey of stroke shows that genetic factors play an important role in the pathogenesis of stroke.In environmental factors,atherosclerosis,hypert-ension,hyperlipidemia,diabetes and heart disease so on are the risk factors of stroke which are associated with heredity.Searching for predisposing genes of stroke is the focal point of the study for genetic mechanism of stroke.Present investigations indicate,p47phox is the functional reconstituent of NAD(P)H oxidase,and acts as a critical role in the activation of the leukocyte oxidase,while reactive oxygen species is significantly associated with the development of ischemic stroke , hypertension and atherosclerosis,and is the risk factor of vascular disease.NAD(P)H oxidase is the principal source of reactive oxygen species in neutrophilic leukocyte and vascular system,and plays an important role in the ischemic lesion and atherogenesis(As).Animal experiment and clinical research show ,reactive oxygen species plays an important role in cerebral ischemia reperfusion,p47phox is associated with ischemia reperfusion injury from p47phox function in ROS generation.It is clear that,if NADPH oxidase subunits coding gene polymorphism or gene mutation lead to increase ROS generation,on the hand,redundant ROS maybe accelerates atherogenesis and the development of hypertension and diabetes by direct oxygenation and indirect acting as second messenger to participate in signal transmission,which lead to increase the susceptibility of stroke;on the other hand,redundant ROS maybe aggravates the lesion of brain tissue by direct injury and indirect acting as second messenger to mediate apoptosis in cerebral ischemia reperfusion. According to the regulation of candidate gene and p47phox function in ROS generation,we presume that p47phox gene is the candidate gene of cerebrovascular disease. As a new tool of studying gene polymorphism, single nucleotide polymorphisms(SNP) is the third genetic marker and have been widely used in identifying susceptibility genes for common diseases.at present,there is no report on the study of relationship between...
