Endogenous antibiotic peptides are widely distributed among various species from plants to mammals. As exhibits a broad spectrum of antimicrobial activities against bacteria, fungi, and viruses, it also named defensin.β- defensins are found to be divided into 28 subtypes. humanβ-defensins 2(hBD-2) play an important role in host defense against bacterial infection in the epithelial barrier, and is the first defensin that is induced upon inflammatory stimulation, and play a unique role in bridging innate and and acquired immune responses.So it exerts huge effects in the lung infectious diseases and skin disorders. In the actual clinical work, Resistance to drug is major difficult problem of using antibiotics.As it has microbicidal activities against bacteria, fungi, and viruses, hBD-2 is researched as a focus.But exploitating of hBD-2 encounters a lot of questions such as synthesizing is expensive and it is not easy to produce through genic program. In this study,we investigate the effect of TNF-α, LPS and pneumococcus on the expression of hBD-2 in the human bronchial epithelial cells(HBECs); and we examined the activity changes of NF-κB by EMSA, and the changes of interacellular calcium by using laser confocal microscopy, and last we investigate the possible role of the intercellular calcium cheater and extracellular calcium chelator and ATP in the TNF-αinduced of hBD-2 gene expression in the HBECs.The objects is explore the possible signal pathway of inducing of hBD-2 , and provide the theoretical basis for induced endogenous hBD-2 expression to against microbial infection.Methods1. Bronchial tissues derived from a lobectomy or pneumectomy from patients ,and bronchias were digested by proteinaseⅩⅣand epithelial cells were scraped off the luminal surface,subsequently cultured in serum-free of DMEM/F12.At last it were indenfied by microscope and cytokeratin(AE1/AE3) of immunocytochemistry.2.
