As one of key important components of HIM,hematopoietic stromal cell has relationship not only with the homing,proliferation,differentiation and self-renew of HSC/HPC,but also with the occurrence,progress and prognosis of some hematologic diseases. So it is very important to study the function of stromal cell on hematopoietic regulation.The cell components in cord blood are abundant. Hematopoietic stem cell in cord blood is more primitive than it is in bone marrow and peripheral blood,and have several virtues , including extensive source , convenient collection , light GVHD and high proliferation. There is great promise for its clinical application. Now, the research on hematopoietic stromal cell mainly focuses on bone marrow stromal cell. Further and systematic study on hUCBSC is badly needed and many questions hasn't been clear,such as,is there any stromal cells in cord blood?how about the biological feature of hUCBSC?if it can be used in the clinic as one of the new resources?etc.VCAM-1 plays a key role in the stromal cell regulated haematogenesis. VCAM-1 can bind with the Integrinα4β1, then strengthen the adhesion between the hematopoietic stem cell and bone marrow stromal cell,and it is very important for the proliferation, differentiation and immigration of HSC/HPC.Based on the background above, in this study, human umbilical cord blood CD34+ cells were seperated and cultured with Dexter system, and the feasibility of the expansion of hUCBSC in vitro and the function of supporting haematogenesis were observed. To up-regulate and stabilize the expression of VCAM-1, an adenovirus vector with VCAM-1 and fluorescin was constructed and transfected to the hUCBSC, then these cells were infused to the athymic mouse whose HIM are damaged by irradiance.
