Choriocarcinoma can be sequential to all of the recognized types of pregnancy. Because of its highly invasive and destructive nature, widespread metastases appear in comparatively early stage, Choriocarcinoma showes a good response to chemotherapy, the utilization of chemotherapy combined with surgical resection, radiotherapy and immunotherapy which has resulted in remission rate of carcinoma greater than 80%. However there were still some patients died, despite initial positive response to chemotherapy. The developments of muitidrug resistance in tumor cells may be a major obstacle to successful chemotherapy .The phenomenon of muitidrug resistance is often associated with increased expression of the mdr1 gene, which encodes P-glycoprotein. Cytokines are proteins produced by the body's immune system, some of which have been shown to modulate muitidrug resistance. However, cytokines administered by the conventional intravenous method can cause severe side effects. Transduction of cytokines genes into tumor cells constitutes an alternative approach for producing and release of the cytokines proteins in the local tumor microenvironment, which may solve problems of toxicity associated with systemic administration.In this study we transfected the human IL-2 gene into the established drug resistant choriocarcinoma cell lines and investigate the reversal of the muitidrug resistance.Methods and ResultThe reseach includes the following two parts: 1. The establishment of the drug resistant choriocarcinoma celllines.Method: We have established 6 resistance cell lines by stepwise or pulse exposure choriocarcinoma cell line JEG-3 to 5-Fu, MTX and VP-16 for 10 months. We analyzed the expression of MDR1 , MRP , LRP , DHFR and GST- π by RT-PCR .determined the chemosensitity of the cell lines to MTX, VP-16, KSM ,TAXOL and 5-Fu by MTT cytotoxcity assay.
