Studies On The Models For Th1 Type Responses And For Exploring The Mechanisms Of Immune Modulation Of Antipsoriatic Drugs

Objectives Psoriasis is a disease with immunologic abnormality characterized by predominance at function of type I helper cells. In order to imitate the feature of the immunological abnormality in psoriasis, some physiopathological processes and links were designed and the possible synergistic effects on the production of Th1 type cytokines EL-12 or IFN-γ were explored. The mononuclear phagocyte system activators LPS or imiquimod were used cooperatively with the T cell mitogen ConA in the culture systems of the spleen cells of Balb/c mice in vitro, and the interfering tests of some anti-psoriatic drugs and probably related compounds were to be performed as well. To aim at establishing some pharmacodynamic experimental models or methods to serve the reevaluation of the known drugs further and seeking the new active compounds in the field of the anti-psoriatic drugs, and/or to serve also the studies on the mechanisms of action of the drugs which possibly regulated the abnormal activation of T helper cells.Methods The spleen cells of Balb/c mice were cultured under the medium RPMI 1640 in vitro with a range of concentrations of ConA combined with the different concentrations of LPS, and moreover, the spleen cells of Balb/c mice fed with 30mg/kg imiquimod once were cultured in vitro with 0.25mg/ml ConA . Then, the levels of the cytokines EL-12, IFN-γ, IL-4, IFN- α were detected with ELISA. On the bases of gaining synergistic effects of cytokines production with ConA 0.25μg/ml and LPS 0.5μg/ml or imiquimod 30mg/kg, the pharmacological activities for immunoloregulation of following drugs or compounds were measured.
