To investigate the bone marrow-derived lymphatic endothelial cells (LECs) contributing to lymphangiogenesis in tumor. We established a method of induction of mononuclear cell from bone marrow to differentiate into LECs in vitro, then to detect the specific markers of LECs such as LYVE-1 and Podoplanin by immuno-fluorescence. We establish a model of tumor lymphangiogenesis and bone marrow derived LECs with tumor-bearing mouse. The VEGF, VEGF-C, b-FGF and Bevacizumab were used to find the effects of the LECs location and mature. The LYVE-1, Podoplanin and Prox-1 mRNA levels were detected by RT-PCR. It is proved that the mononuclear cell from bone marrow could be induced to differentiate to LECs successfully in vitro, and some new LECs derived form bone marrow.
