| Objective : To investigate the role of Tumor necrosis factor-α(TNF-α) and Tumor necrosis factor receptor Ⅱ (TNFR Ⅱ) in preterm labor and their gene polymorphisms in genetic susceptibility to preterm labor in Chengdu. Methods: We collected 46 cases maternal serum and partial placental tissues of preterm labor(21 cases of infectious proup with chorioamnionitis, 25 cases of noninfectious group without chorioamnionitis), and 50 cases maternal serum and 20cases placental tissues of term labor in corresponding period. (1) TNF-α and TNFR Ⅱ mRNA were tested in 66 cases placental tissue by reverse transcription polymerase chain reaction(RT-PCR). (2) 66 cases serum levels of TNF-α and sTNFR Ⅱ were measured by enzyme-linked immunosorbent assay(ELISA). (3)SP/SABC immunohistochemical staining was employed to detect the location and quantity of TNF-α and TNFR Ⅱ in 66 cases placentas. (4)Using polymerase chain reaction-restriction fragment length polymorphisms (PCR-RFLP) methodology, TNF-α and... |
