Objective (1) To investigate the effects of 15d-PGJ2 (15-deoxy-A12,14-PGJ2)at orthotopic liver transplantation in rats (ROLT); (2) To judge the effect mechanism of 15d-PGJ2 in ROLT. Methods (1) We divided the rats into 3 groups after we have built the ROLT model successfully by two-cuff method: non-rejection groups(LEW→LEW, group A), rejection groups(DA→LEW, group B),control groups(DA→LEW +PGJ2, group C);13 rats in each group.8 rats were killed at the 14th postoperative day, others were kept to observe the survival . (2) To observe the symptoms and pathologic changes, the plasma levels of the liver function(TBIL,AST ALT,ALB);(3) To detect the PPARγmRNA in liver by real time-PCR, cytokine(IL-2 , IL-10) levels in plasma were also measured with enzyme linked immunosorben assay; (4) To examine the proliferation activity of T lymphocyte in spleen by MTT assay,and the apoptosis in liver lymphocytes by TUNEL.Results (1) Duration of operation was not significantly different among 3 groups.
