ObjectiveTo observe expression of μ-calpain gene and protein, caspase-3 activation and apoptotic cells in neonatal rats after hypoxic-ischemic brain damage and to investigate the effect of calpain inhibitor-3(MDL28170)on the brain after HIBD. MethodHypoxic-ischemic brain damage model was built up using neonatal rats. Cerebral pathological and ultrastructural changes were observed and CAl neuronal loss was counted. Quantitative real-time flurogenic polymerase chain reaction was employed to detect μ-calpain gene expression, western blotting technique was used to detect μ-calpain protein activation. Caspase-3 activation was measured by immunoblotting and immunohistochemistry. Apoptosis was detected by terminal deoxynucleotidyl transferase mediated d-UTP nick end labeling staining(TUNEL).
