After isolated culture in vitro, BMSCs can different into many kinds of cells, such as osteoblast; cartilage cell; adipocyte; cadiocyte; nerve cell, in some induce condition. It can not only be induced and different iateds into various kinds of neurocyte and neuroglia cell but also protect and nutrition cells and tissue by secrete many kinds of cytokines. Now, it has been found that SDF - 1/ CXCR4 play a very important role in stem cell mobilization, migration, and homing. CXCR4 can express in haemopoietic stem cell(HBSC), CD34 + cells. The chemotatic factor released by tissue can make effect by CXCR4 and adhesion molecule on surface of cells, in final to guide stem cells homing successfully, at the same time, to assist stem cell spanning endothelial cell. In many condition , such as tissue ischemic or cell damage, cells can secret SDF - 1, inducing stem cell migrate to injury tissue. It can release a great quantity of cell factor after SCI, such as interleukin, SCF, GM - CSF, C - CSF, M - CSF, SDF - 1, etc. they could take effect in migration of BMSCs. It can be certain that haemopoietic stem cell express CXCR4, but whether it express in BMSCs has been disputed. It need be further studied what role SDF - 1/CXCR4 is in the migration of BMSCs. on account of the result and problem above - mentioned, this study views from the angle of expression of CXCR4 in BMSCs, verify the effect of SDF - 1 promoting the migration of BMSCs, In vivo, observe this kind of aid in SCI.Meaterials and MethodsHarvesting cells from femur and tibia medullary canal by operation. Separate and purify these cells utilizing the characteristics of adherence of BMSCs,...
