Shrimp is one of the best animal proteins for human; it is also one of the eight kinds of foods that frequently causes allergy. As the development of world trade, shrimp was used more and more widely in all kinds of foods. The potential risk of shrimp allergy was worth to pay more attention. This paper mainly focused on shrimp allergen separation and purification, polyclonal antibody preparation, immunodetection, some factors that affect shrimp allergenicity and diagnose of shrimp allergy. The results are as follows:1. Sera from different patients can identify different shrimp proteins. In the experiments, there are 8 proteins was recognized. Among these, more than 80% patients recognized one of 36kD protein, which was called tropomyosin; the second one was 85kD protein; apart from this, some patients also recognized 52kD, 41kD, 31kD, 30kD, and 22kD proteins. As for different species of shrimp, although protein extracts in SDS-PAGE was different slightly, the major shrimp allergen was identical, the allergenicity of different species was not obvious different.2. In the paper, Dot-ELISA system was used to detect shrimp allergen immunoreactivity. Using purified shrimp allergen as antigen, BALB/C mice and New Zealand rabbit was used as animal to product polyclonal antibodies, the value was 320,000 and 160,000 respectively. The author used S180 to stimulate mice to produce ascites fluid, the result suggest that there are high value (80,000) antibody in the ascites fluid, it is an effect way to produce polyclonal antibody. After purification, the value was double to the original sera. Western blotting results suggest that it is special to shrimp allergen.4. Using rat antibody, Ci-ELISA methods was used to detect shrimp allergen, LOD was 1.85ng/mL, linear lest detection was 4.79ng/mL-1400ng/mL, CV in vivo plate was 3.4%-9.1%; CV in vitro plate was 12.9%-19.6%. The kit can reach the level of the product in the world.
