Part 1 Effect of 1-methylhydantoin on HK-2 cells apoptosisand expression of fibrotic correlation factors andintervention of medicineObjective:To investigate the effect of 1-methylhydantoin on cell cytotoxicity, apoptosis and mRNA expression of some cytokines in human renal tubular epithelial cell line (HK-2),and the intervention of PGE1 and probucol to them.Methods: (1)Human PTEC cell line HK-2 was used as the subject. (2)The cell inhibitory rate of HK-2 was detected by MTT method.(3) The cytotoxicity of 1-methylhydantoin to HK-2 was determined by NAG release test. (4)Cell apoptosis was evaluated by using Hoechst stain and FACS with Annexin-V/PI. (5)Immunocytochemistry , Flow cytometry and RT-PCR, were employed to investigate the follows:the protein and mRNA expression of TGF-β1,CTGF and FN. In the blocking study , PGE1 and probucol were used as antagonists respectively. The changes of TGF-β1,CTGF and FN level and apoptosis were compared.Results: The OD value and NAG concentration of creatinine, 1-methylhydantoi compared with that of normal control , OD value decreased and NAG concentration increased significantly (P<0.01).when...
