Expression And Cellular Localization Of A Novel Cerebral Ischemic Pretreatment Related Gene Mipu1 And Identification Of Mouse Mipu1 Gene Homology

Part 1 Global cerebral ischemic pretreatment and ischemia/reperfusion model of rat.Objective To establish global cerebral ischemic pretreatment and ischemia/reperfusion model of rat and to investigate the protective effects of cerebral ischemic pretreatment on the following cerebral ischemia/reperfusion injuries. Methods 48 SD rats were divided into 2 groups randomly: experimental group and sham-operated control group. The cerebral ischemic pretreatment and cerebral ischemia/reperfusion model were established by ameliorated four-vessel occlusion. The experimental group was treated with cerebral ischemic pretreatment for 3 minutes to compare with sham-operated control. Different time points after operation two groups were all treated with 6 minutes cerebral ischemia followed by 24 hours reperfusion,then examined by detecting brain water content or observing histological change. Results Compared with the sham-operated control group,the brain water content and histological change of experimental group which was treated with ischemia/reperfusion 24 hours after pretreatment were reduced. Conclusion Global cerebral ischemic pretreatment in rat can induce protective effects on ischemia/reperfusion injuries 24 hours after the pretreatment.Part 2 The expression of Mipul gene in the brain of rat after cerebral ischemic pretreatment.Objective To investigate the expression of Mipul gene in hippocampus and cortex of rat brain at different time-points after ischemic pretreatment.
