The clinical outcomes of hepatitis B virus (HBV) infection are extremely various, and might be associated with host genetic factors, viral factors and environmental factors. A total of 212 HBV self-limited infection individuals (self-limited individuals), 244 asymptomatic HBV carriers (HBV carriers) and 391 chronic hepatitis B (HB) patients (HB patients) were recruited to conduct a case-control study. TNF-α-238G/A, -857C/T, -863A/C and VDR-Fok I T/C, Taq I C/T gene polymorphisms were examined by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP), and HBV genotypes were examined by nested PCR. The results related to the association analyses of TNF-α gene, VDR gene, HBV genotype and drinking with outcomes of HBV infection, and the analyses of interaction between gene-gene and interaction between gene-environment are as follows:1. The frequencies of TNF-α-238 genotype GG and GA in chronic HB patients were 90.0% and 10.0%, respectively, and 89.3% for genotype GG and 10.7% for genotype GA in HBV carriers, and 94.8% and 5.2% for genotype GG and GA in self-limited HBV infection individuals. There was no statistically significant difference in the distribution of -238G/A genotype between HB patients and HBV carriers (x~2= 0.08, P=0.78), and statistically significant differences were founded between self-limited individuals and HB patients, and between self-limited individuals and HBV carrier (x~2= 4.13, P=0.04; x~2= 4.55, P=0.03). It was found that self-limited individuals with TNF-α-238 GA genotype had a 2.02-fold increased risk of chronic HB (95% CI: 1.01-4.04; x~2=4.13, P =0.04), and had a 2.18-fold increase risk of HBV carriage (95% CI: 1.05-4.52; x~2=4.13, P =0.04). A multiple logistic regression analysis also demonstrated that TNF-α-238 GA genotype was independently associated with HBV carriage (OR=2.34, 95% CI: 1.12-4.89), and was independently associated with chronic HB (OR=2.11,95% CI: 1.02-4.38), after adjustment for sex and age.The frequencies of TNF-α-857 genotype CC, CT and TT in HB patients were 79.8%,...
