Epidermal Stem Cells (ESCs) are important resource of skin development, metabolism and wound healing. ESCs not only control cuticularization and metabolism, but also participate in hair-follicle morphogenesia, cycle of hair follicle and repair of follicle damage. Therefore, to illuminate the regulation mechanism of differentiation of ESC will benefit to find new treatment measures for promoting wound healing and improving wound-healing quality.Previous researches have verificated that P-catenin expressed in ESCs is a key protein to switch on differentiation of ESCs and determine direction of ESCs differentiation. As an important glucoprotein of Wnt-signaling pathway, even low level of P-catenin is necessary for ESCs to maintain stem cell state. Once its expression is up-regulated, ESCs will enter differentiation procedure. ESCs will develope toward hair follicle followed high expression ofp-catenin, but will developed toward epidermis cells with removing P-catenin. Hence it is very important to illuminate regulation mechanism of P-catenin expression with an aim of controlling direction of ESCs differentiation, promoting follicle repair or reconstruction of follicle.Sensory neuropeptide substance P (substance P, SP) has been studied for many years as a cytokine. Previous studies were generally focused on its effectiveness acted as neurotransmitter, hormone and modulator. SP was not only releases from sensory nerve ending after wounding, but from fibroblast and endotheliocyte, which can up-regulate expression of SP gene and proteion, promote cell proliferation and induce some cytokines and proteins related with wound-healing(EGF, bFGF, β-catenin, VEGF, MMPs, NF-κB fibronectin et al) to be expressed.It has been noticed by our previous researches that SP is an important cytokine for controlling differentiation and migration of ESCs.
