Evidence provided by molecular biology studies showed that cytokine may play an important role in the autoimmune reaction of Sjogren' Syndrome. Efforts were made to understand the mechanisms of the cytokine in the inflammatory process at molecular biology level. In this dissertation, we investigated the expression of cytokines in labial gland biopsies and also the effect of human rTNFα on outgrowth epithelial cell of labial gland in vitro.In assuring the specificity of hybridization in situ in detecting cytokins mRNA, we compared hybridization in situ using TNF α cDNA probe labeled with biotin and immunohistochemical staining with TNF α monoclonal antibody on consecutive labial gland sections. The result showed that the distribution of TNF a mRNA and TNF a molecule were completely consistent in the acini of labial gland. Also dot blot hybridization confirmed the sensitivity of the test. All these implied hybridization in situ is a reliable test in detecting local CK expression.To investigate the coexpression of cytokins, we established in situ non-isotope double hybridization method , which allowed mRNA of two cytokines coexpressed in one single cell to be seen under the light microscope.
