Celastraceae plants have been the subject of continued and growing interest, due to the range of biological activities shown by many members of this family. The pharmaceutical studies and clinical practice have demonstrated that its sesquiterpenes and tnterpenes possess notable antibacterial, anti-tumor, insect antifeedant and cytotoxic activities.More than 100 compounds were isolated, purified from three species Celastrus hypoleucus (Oliv.) Warb., Celastrus rosthornianus Loes., and Microtropis triflora Merr. et Freem. belonging to the Celastraceae family. The structure was determined by IR, UV, MS, NMR and X-ray diffraction analysis, respectively, and thirteen of them are new compounds isolated from plants.1. The chemical study on methanol extracts of the stalk of Celastrus hypoleucus (Oliv.) Warb. leading the isolation and identification of five new compounds together with thirty-seven known compounds. Their structures were elucidated on the basis of spectral and chemical evidence, respectively, and especially, by X-ray diffraction analysis for compound A-4. The five new compounds were determined as follows: 6α-hydroxy-12-oleanen-3-one (A-4), 12-oleanen-3β, 6α-diol (A-5), 12-oleanen-3β, 23-diol (A-6), (+) 7-deoxynimbidiol (A-28), and 12, 13-Dihydroxy-3, 8, 11,13- celastratrtraen-7-one (A-29).2. Forty compounds were isolated from methanol extracts of the stalks of Celastrus rosthornianus Loes. and their structure were identified by analysis of spectral data, respectively. Among them, six new natural compounds were elucidated as: (3β, 20(S), 24(S)-trihydroxyldammar-25-ene-3-caffeate (B-1), 3β, 20(S), 24(R)- trihydroxyl-dammar-25-ene-3-caffeate (B-2), 3β, 20(S), 25-trihydroxyldammar- 23(Z) -ene-3-caffeate (B-3), 1β, 3β, 11β-trihydroxy-olean-12-enyl-3-palmitate (B-12), 1β, 3β-dihydroxy-olean-9(11),12-dienyl-3-palmitate (B-13), and 1β-acetyloxy-4α-hydroxy-6α, 9α-bis(benzoyloxy)-β-dihydroagarofuran (B-24).3. Twenty-five compounds were isolated and identified from methanol extracts of the stalks of Microtropis triflora Merr. et Freem.. Among them, 2α, 3β, 21β-trihydroxy-22-oxo-29-nor-24, 2/?-friedelanolide (T-12) and 2/?-hydroxy-24, 29-nor-4-friedelene-3, 21-dione (T-13), named triflora lactone and triptocalline B, respectively, were new friedelane type triterpenes.In vitro test (MTT method), new compounds A-5 and A-28 showed moderate anti-tumor activities against human mammary carcinoma (Bcap37), human colon carcinoma (RKO), human hepatocellular carcinoma (SMMC7721) and human erythroleukemia (K562). Compounds A-4, A-29, B-l, B-2, B-3, T-12, and T-13 showed anti-tumor activities against human cervical squamous carcinoma cell (Hela).In addition, the advances in isolation and bioactivities of components from Celastraceae were reviewed.
