To study the expressions of sFRP, β-caternin, Cox-2, ER-β, pS2, Keratin 19 proteins , Microsatellite Instability (MSI) gene in colorectal adenocarcinoma cells or in local lymph nodes micrometastasis cells of colorectal adenocarcinoma and their correlations with prognosis of colonectal adenocarcinoma, the investigations were carried out by using the methods of Tissue Microarray, immunochistochemical staining, fluorescence PCR (F-SSCP).The results showed that: 1) the expressions of SFRP4 and β-catenin proteins in colorecta carcinoma were significantly high than in normal colorectal mucosa and β-catenin was significantly high in P53 and hMLH1 positives; 2) the expression of pS2 was signifieantly high in colorectal carcinoma. In 90 cases for MSI positive, the positive immunohistochemical stanning was significantly high (45.5%) compared to in 801 cases for MSI negative (30.0%). No correlation between overexpression and prognosis and survival in colorectal carcinoma after surgical excision. 3) the expression of kenatin 19 was strongly positive in mesorectum lymph nodes. The expression of keratin 19 in tumor cells was higher in Dukes C/D than in Dukas A/B. In 25 eases for HE negative staining, the strong immunohistochemical positive staining was observed in 9 patients. However, no relationship between theexprecsion of Keratin 19 and the age of the patients.In conclusion: 1) no relations with the expressions of SFRP4 and β-catenin and stages, grade and prognosis in colorectal carcinoma. SFRP4 was likely to be a upregulate gene in colorectal carcinoma; 2) the expression of pS2 was significanlly high in the deficience of mismarched repairing genes (P53 and hMLH1). No corelations between the overexpression of pS2 and prognosis in patients got colorectal carcinoma; 3) the immimohistochemical staining was more sensitive in lymph nodes micrometastastic cells than that of HE staining. The expression of Keratin 19 in Dukes C/D was significantly higher than in Dukes A/B. Moreover, for the patients with micrometastases in local lymph nodes had a poor prognosis than the patients with no micrometastases.
