Surgery, radiation-theraphy and chemotherapy have played an important role in the cancer treatment of head and neck. But the multidrug-resistance is a kind of special phenomenon that is characterized by drug-resistance of tumor to various drugs. In clinic, mucoepidermoid carcinoma is traditionally taken for the tumor that tends to resist chemotherapy. However, its mechanism is rarely investigated and left unclear. The aim of this study is to explore the related factors of multidrug-resistance in mucoepidermoid carcinoma, to detect its mechanism at protein and gene levels, thereby, assisting in guidance of choosing chemotherapeutics protocols and enhancing the effect of anticancer drugs.The study is composed of three major parts, establishment of multidrug-resistant model of Mc3 cells by drug inducement in vitro with its detection and analysis; establishment of multidrug-resistant model of transplanted mucoepidermoid carcinoma in nude mice by drug inducement in vivo with its detection and analysis; the investigation of the gene expression profiles to the multidrug-resistant model of transplanted mucoepidermoid carcinoma in nude mice by gene chip.The results show that the multidrug-resistant model of Mc3 cells has beenestablished by drug-inducement in vitro. The Mc3/5-Fu cells behave the multidrug-resistance characteristic; the multidrug-resistant model of transplanted mucoepidermoid carcinoma in nude mice has been established for the first time by drug-inducement in vivo; the Mc3/5-Fu cells resist many drugs, not only the 5-Fu, the inducing drug, but also other drugs like MTX, PYM, VCR. This phenomenon agrees with the cross-resistance phenomenon in clinic; P-gp and GSH-GST have been found related to multidrug-resistant mechanism of mucoepidermoid carcinoma by immunohistochemistry, RT-PCR and gene chip, in cells and in tissue; P-gp and GSH-GST might be the potential mechanism of initial drug-resistant in mucoepidermoid carcinoma; the multidrug-resistant mechanism of mucoepidermoid carcinoma might relate to DNA- topoisomerase, HSP-HSF, apoptosis and cell cycle regulation.The study estimates the multidrug-resistant mechanism of mucoepidermoid carcinoma, comparatively roundly, for the first time. These results would lay a foundation to go deep into the detail and to its reverse.
