PURPOSE: The expression of Flk-l, LRP and MDRl gene protein were examined immunohistochemically in 70 primary lung tumors (59NSCLCs and 11 SCLCs) and 5 normal lung tissues.The relationship was investigated between the results and clinic and pathologic parameters, especially the clinical outcome in non-small cell lung carcinoma(NSCLC). The aim was to find some valuable criteria for diagnosis, prognosis and treatment of the lung cancer.RESULTS: Of the 70 lung cancers, 29 cases (49.2%) were positive for MDRl expression in NSCLCs and 2cases (18.2%) in small cell lung carcinoma(SCLC); 41eases (69.5%) were overexpression for LRP in NSCLCs and 3 cases (27.3%) in SCLCs, There were statistically significant correlation (P<0.05). The squamous carcinoma was significantly different(P<0.05) from adenocarcinoma in the positive rate of MDRl and LRP. The co-expression rate of LRP with Flk-1 in 59 cases NSCLCs was 49.2%. There was significant correlation between the expression of LRP and histological grade or smoke (P < 0.05). There was statistically significant correlation between the expression of MDRl , LRP and histological type, the expression of Flk-1 and TNM staging also (P < 0.05). Flk-K MDRl and LRP genes showed co-expression in some of the lung tumors. In a multivariate analysis by Cox regression model, the overexpression of Flk-1+LRP , Flk-1+LRP+ MDRl , the traditional Chinese Medical therapy and recrudescence were independently associated with patient survival length (P<0.05).CONCLUSIONS:The experiment suggests that Flk-K MDRl andLRP genes play roles in oncogenesis > progression and therapy of lung cancer.Detection of these geneproteins has positive effects on diagnosis prognosis and treatment of lung tumors.
