Objective:To investigate the mechanisms of Electroacupuncture on apoptotic genes in Alzheimer's Disease (AD) , learning and memory ability , resisting the oxidative stress on AD rats, and to explore the changes of apoptosis of nerve cell and the apoptotic genes in AD rats induced by unilateral amyloid β 25-35 protein(A β 25-35).Methods:Experimental rats were randomly divided into model control group, model group, Electroacupuncture-treatment group and aFGF-treatment group. AD model was induced by unilateral amyloid β 25-35 protein(A β 25-35) injected into the lateral ventricle of rats. The points of baihui,dazhui,taixi and zhuosanli were chosen to practise Electroacupuncture. The following techniques were used to observe the effects of Electroacupuncture on Leaning and Memory Impairmernt, cell apoptosis ,the expression of apoptotic gene and resisting of the oxidative stress induced by A β 25-35 in rats: The Morris water maze was used to detect the visual spatial learning and memory ability of every rat. Contents of MDA, H2O2 and enzyme activity of CAT were measured by diagnostic Kit. Protein and mRNA expression of p53, Bax, bcl-2 were detected by Western-blot and RT-PCR respectively. Immunohistochemistry for analysis of the expression of gene related with Hippocampus apoptosis. HE staining for observing histological features of apoptotic cells in brain.Result:1.
