Effect Of Niaosuanping On Experimental Hype Ruricacidemia Rat

Abstract/Summary:
To observe the effect and to explore the mechanism, we give the experimental hyperuricaci-demia rat with Niaosuanping apozema for curative and protective research.1. To make the animal model of hyperuricemia associated with uratic nephrotic syndromeAccording to the content of serum uric acid, we make SD hyperuricemia model with different doses of adenine (50mg/kg, 100mg/kg, 150mg/kg). After fed SD rats with adenine for 32 days, we change to common forage. Then divide them into subgroup according-to the content of serum uric acid again. To prove the model has been made, we treat them with allopurinol (53.57mg/kg) for 16days, after that, we observe the level of serum uric acid successively and test the renal function.(1) High and middle dosage adenine increase the content of serum uric acid (P<0.01). After treatment with allopurinol, the contents of serum uric acid decrease (p<0.05).(2) High and middle dosage adenine increase the content of serum uria nitrogen and creatinine (P<0.01). After treatment with allopurinol, the levels of serum uria nitrogen and creatinine keep high (P>0.05).(3) Swelling renal tubules, partial deposition of crystalline in cavities, infiltration of inflammatory cells and proliferation of interstitial component appear in high and middle dosage adenine groups; Under electron microscope, we find microvilli of the renal tubules epithelial cells dissolved, lysosomes increased, basement membrane of renal corpuscle thickened and partially confluenced. Among them, the curative model has severer inflammatory damage compared with contrast group.2. The curative effect of Niaosuanping apozema on hyperuricemia animal model Based on the first test, we make model with 100mg/kg adenine. After 5 weeks, wedivide the rats into 3 groups, according to the content of serum uric acid, and feed them with different dosages of Niaosuanping apozema (3.05g/kg, 6.10g/kg, 12.2g/kg) for 48 days; Allopurinol is the contrast medicine.(1) High and middle dosage Niaosuanping apozema decrease the content of serum uric acid (P<0.05), but slower than allopurinol group.(2) Niaosuanping apozema can lower the active of XOD (P<0.01), but slower than allopurinol group.(3) Niaosuanping apozema can decrease the content of serum creatinine (P<0.05). Among them, the effect of middle dosage group appear fastest. All the medical groups have increased clearance rate of creatinine (P<0.01); Low and middle dosage Niaosuanping apozema decrease the content of serum uria nitrogen (p<0.01), but the high dosage Niaosuanping apozema and allopurinol can not; Niaosuanping apozema can increase the content of uric acid in urine (P<0.01), but the results is opposite in allopurinol group (P<0.001).(4) In middle dosage Niaosuanping apozema group, the form of renal tubules, renal corpuscle and interstitial structure are intact; Crystalline deposited in renal medulla, infiltration of inflammatory cells in interstitial component and various degrees of proliferation appear in model contrast group, allopurinol group and low dosage Niaosuanping apozema group. The form of partial cells in high dosage Niaosuanping apozema group is abnormal and the interstitial component is swelling. Under electron microscope, we find the renal tubules epithelial cells in middle dosage Niaosuanping apozema group are nearly normal, whereas those in other groups are pathologic.3. The protective effect of Niaosuanping apozema on hyperuricemia animal model Feed SD rats with different doses of Niaosuanping apozema (3.05g/kg, 6.10g/kg,12.2g/kg) and 100mg/kg adenine for 48 days simulanously; allopurinol is the contrastdrug.(1) Middle or high dosage of Niaosuanping apozema and allopurinol all can depress the increasement of serum uric acid and tha activity of XOD, while the low dosage group depresses slowlier (P<0.001).(2) Middle or high dosage of Niaosuanping apozema and allopurinol all can depress the increasement of serum creatinine, while the low dosage group depresses slowly (P<0.001); middle dosage group have increased clearance rate of creatinine, while other groups works slowly (P<0.001); Middle or high dosage of Niaosuanping apozema all can depress the increasement of serum uria nitrogen, while the low dosage and allopurinol group depress slowly (p<0.001); Compared with allopurinol group, Niaosuanping apozema can increase the content of uric acid in urine (P<0.05).
