| Primary hepatic carcinoma is a kind of highly malignant tumor of digstive system which has highly incidence and mortality in our country. Now combined therapy including operation, radiation therapy , chemotherapy and biotherapy are clinically used to deal with it.However, there are still some difficult problems such as highly recurrence, strong side effects and bad therapeutic effects on mid-and advanced stage of this disease. In this study,we detect the inhibition of metabasis and proliferation of Human Hepatocarcinoma in Intense Magnetic Field going in for finding a safe, efficacious and convenient type of new aided therapy.Method: The Intense Magnetic Field is constructed by a shape of U pure Ferrum.The two Magnetic poles inside the Ferrum was composed of two layers of Neodymium -Ferroboron which was magnetized in advance. The cross Section of Magnetic poles is 200×200mm and the distance between the two Magnetic poles.is 110mm. The intensity of magnetism on the pole surface is 0.6T and 0.30-0.35T in the middle distance. The Grad of intensity of magnetism is 0.55T/m. Such kind of designation forms inside- magnetic circuit which efficiently reduce the damage of magnetism intensity between the two poles. In vitro experiment: SMMC 7721 was divided in experimental groupand control group. The experimental group were composed of 3 groups,8-day(El),16-day(E2) and 24-day(E3) exposure to the Intense Magnetic Field.Results:The growth curve of E1,E2 and E3 are lower than the Control group and cell inhibition rate was 32.3% , 68.7% , 72.1%, dividely.There is significant difference between El and E2, (P<0.01) in other to E2 andE3 (P>0.05) ;The adhesive power and cell movement power of E1 ,E2 and E3 are both lower than the Control group (PO.05, PO.01, P<0.01) ;Cells in S stage of El and E2 are more than Control group. (P<0.01) . The Apoptosis rate of E2 was 10.78%.Morphology: Under light microscope,the cells are inhibited obviously and in loosen arrangement.There is not much cells in splitting stages and less heterotype cell nucleus. Under transmission electron microscope,there were chronic and retrogressive injury in cells, the cell organs was unclear and much myelinfigure indicated that retrogressive changes. The karyoplasmic ratio are lower than control group.Animal experiment: 40 cases of nude mice were divided into 4 groups.The mass of Tumor which has been passaged in nude mouce for 137 generation was innoculated in left hepatic lobe and fore-leg oxtar.All these high- metabasis of human liver cancer models were divided into Group I (8-hour exposure / d) ; Group II (4-hour exposure / d) ; Group III (2-hour exposure / d);IV Control Group. All groups are exposured to The Intense Magnetic Field 3 days after innoculation. Remove subcutaneoulytumor after 24-day exposured to The Intense Magnetic Field. Live time and survival rate of all the mice are recorded, tumors in liver and metabasis are recorded,too. Histological section of tumor ,liver and lung dyeing by HE were detected under light microscope. Apoptosis rate was detected by Tunel dyeing method.Result: Survival rate of I group is obviously longer than other three groups and its Apoptosis rate is higher than others in 24-day exposure and more higher in 50-day exposure, metabasis Only occurs in 60 percent mice in I group, Group II has the simlar survival rate with Group III but longer than Group IVConclusion: Intensity magnetic field has inhibiting effects on metabasis and proliferation of Human Hepatocarcinoma SMMC 7721 Cell lines and , the high- metabasis of human liver cancer mice models.This effects can be intensified by longer time exposure to magnetic field.Clinical Case : male, 55years old, 5 years after resection of huge mass liver cancer in Left lobe , had disseminated metastasis in liver,double lungs , subcutaneouly tisues and lumbar vertebra. Therapy: thorax and abdominal organs were completedly exposed to the The Intense Magnetic Field(0.3-0.6T),8h/d, 2 months course of treatmento Result: The patient had better appetite , consciousness and more 5 Kg weight than before,less bone ache ,then reduced doses of Morphina .His WBC rise from 3.5 xio9/L to 4.2xlO9/L, CT SCAN did not indicated more metastasis then before. |
