Objective: In order to discover etiology of Allergic Rhinitis(AR) at gene level, gene chips were used in expression profile study, and gene change with SP and steroid treatment in animal model were observed.Methods: The total RNAs were isolated from mucosa of AR and normal,then purified to mRNAs and reversely transcribed to hybridization probes.Two types gene chips one contain immune genes and the other contain genes of chemokines and their receptors were used in AR diference genes scan.Guinea pig sensitied by ovalbumin for model made and challenged with substance P(SP) were second group,and the last two groups were with steroid treatment and contral. Praxiology markers, IgE, histamine and SP were detected .Gene chip of rat were used in observing genes expression change.Results:Lots of genes concern as immunoloregulation, signal conduct, surface antigen of lymphocytes, cytokines and receptors were found different expressed in profile and highly expressed genes of chemokines and receptors were inclined to Th2. Immunology marks were found higher in AR model than contral,and more higher when SP challenge,but dropped down when steroid treatment.Genes of Sp and its receptor, surface CD antigen, IgE, cytokine and its receptor were found up-regulated in AR and more inflammation genes up-regulated with SP challenge,and most of genes expression were suppressed with steroid treatment. Conclusion:Many genes involve in mechanisms of AR,SP and steroid influence AR at gene level.
