OBJECTIVETo study the effects calcitonin gene related peptide ( CGRP) and nerve growth factor ( NGF) on the expression of c - fos mRNA/c - Fos protein , c -jun mRNA /c - Jun protein , Caspase - 3 protein in striate cortex of rats.MATERIALS AND METHODSOn the model of transient whole cerebral ischemia/reperfusion induced by clamping the bilateral common carotid arteries in rats, the method of in situ hybridization and microscope image analyse were used to observe the expression of c - fos mRNA and c - jun mRNA and the methods of immunohistochemistry and microscope image analyse were used to observe the expression of c - Fos protein , c - Jun protein , Caspase - 3 protein in striate cortex of rats with whole cerebral ischemia.Male SD rats of weight 300 - 350g supplied by experimental animal center of china medical university. Male SD rats were divided randomly into sham group, reperfusion after ischemia (I/R) group,NGF group , CGRP group and NGF plus CGRP group(NGF&CGRP). there are 6 animals in one group. In I/ ; R group, 1ml of 0.9% sodium chloride was injected into internal carotid artery at the beginning of reperfusion after ischemia 30 min. In NGF group, l000uNGF/lml was injected into internal carotid artery at the beginning of reperfusion after ischemia 30 min. In CGRP group, 1 μgCGRP/ 1ml was injected into internal carotid artery at the beginning of reperfusion after ischemia 30 min,In NGF&CGRP group, l000uNGF plus 1 μgCGRP /1ml was injected into internal carotid artery at the beginning of reperfusion after ischemia 30 min,whole cerebral ischemia in rats were induced by clamping the bilateral common carotid arteries and reperfusion began after removing the artery clamps, the bilateral common carotid arteries in rats were just clamped. In sham group,the skin was dissected and the bilateral common carotid arteries in rats were separated only. The animals of deffrent group were executed by decollation at 3h , Id , 3d after ischemia/reperfusion.RESULTS AND DISCUSSION1. The expression of c - fos mRNA in ischemia/reperfusion (I/R) group was increased than in sham group ( p <0. 01). the expression of c - fos mRNA was lower in CGRP group than in I/R group (p < 0. 05 ) , and lower in NGF group than in I/R group (p <0.05). and markedly lower in NGF&CGRP group than in I/R group (p <0. 01) , and lower in NGF&CGRP group than in NGF group and CGRP group(p < 0. 05 ) .2. The expression of c -Fos protein in ischemia/reperfusion (I/R) group was increased than in sham group ( p < 0. 01). the expression of c - Fos was lower in CGRP group than in I/R group (p <0. 05). and lower in NGF group than in I/R group (p <0.05). and markedly lower in NGF&CGRP group than in I/R group (p <0.01) , and lower in NGF&CGRP group than in NGF group and CGRP group(p <0.05 ) . The expression of c - Fos in I/R group was stronger 3h survival after ischemia/reperfusion than Id survival and 3d survival (p < 0. 05) ,and stronger in NGF&CGRP group 3h survival after ischemia/reperfusion than Id survival and 3d survival (p <0.05).3. The expression of c -jun mRNA in ischemia/reperfusion(I/R) group was increased than in sham group (p <0. 01). the expression of c -jun mRNA was lower in CGRP group than in I/R group ( p < 0. 05 ) , and lower in NGF group than in I/R group ( p < 0.05 ). and markedly lower in NGF&CGRP group than in I/R group (p <0. 01) , and lower in NGF&CGRP group than in NGF group and CGRP group( p <0.05).4. The expression of c -Jun protein in ischemia/reperfusion (I/R) group was increased than in sham group ( p < 0. 01). the expression of c - Jun waslower in CGRP group than in I/R group ( p <0. 05). and lower in NGF group than in I/R group (p <0.05). and markedly lower in NGF&CGRP group than in I/R group ( p <0. 01) , and lower in NGF&CGRP group than in NGF group and CGRP group ( p <0.05).
