| The article is to discuss the mechanism of protecting brain and promoting neuroplasticity with acupuncture through the methods of improving resorcinol reagent and high-performance capillary electrophoresis on gangliosides and its contents of GM1, GM3, GD3 in the rat of acute cerebral ischemia. It may provide the basis theory for the clinic therapy. Gangliosides can promote neuroplasticity. Extraneous gangliosides can pass the blood-brain barrier and embed into neuron cellular membrances, exerting the functions of endogenous gangliosides. As indicated by researches, the contents of gangliosides and its acid components GM1, GM3 ,GD3 decreased along with the prolongation of ischemic. There is auto-regulated mechanism of increasing gangliosides compond to alleviate cerebral ischemic, but the auto-regulated is limited. The subject may clarify the relation between the functions of acupuncture and gangliosides. It has discussed the acupuncture mechanism' of promoting nuroplasticty through ameliorating the contents of endogenous gangliosides.204 Wistar rats were randomly allocated into control (n=10), model (n=64), acupuncture (n=64), and medication (n=66) groups. Left middle cerebral artery occlusion (MCAO) of rats in the latter three groups was made by thread-ligation method. And acupuncture treatment was applied 5 minutes after embolism in acupuncture group. The rats of the medication group were injected intraperit -oneally with GM1 (10 mg/kg body mass). Gangliosides content in the brain were measured lh,2h,3h,5h,7h,12h,24h following cerebral ischemia. The content of Lipid-bound Sialic Acid (LBSA, represent GLS) decreased significantly after the acute cerebral ischemia, that of acupuncture group and medicine group increased significantly, two groups,were similar in curative effect , and had difference only at 2h, 3h, 5h, that of acupuncture is lower. Acupuncture has a regulative effect on gangliosides to cerebral protection andneuroplasticity. The content of GM1 and GD3 decreased significantly after the acute cerebral ischemia with time, the content of GM3 had no difference with that of the normal, that of acupuncture group and medicine group increased significantly, and the content of GM3 decreased, GD3 increased significantly in acupuncture group. The results show that acupuncture has a regulative effect on gangliosides GM1, GM3, GD3 to cerebral protection and neuroplasticity. |
