Objective Radix Paeoniae Bubra is the dry root of Paeonia lactiflora pall that is Japanese buttercup herbaceous plant.The drug is bitter in flavour,slightly cold in property,acting on the liver and spleen channels. It has many effects,such as: removing pathogenic heat from the blood,dissipating blood stasis and relieving pain. Preventive effect and mechanism of Radix Paeoniae Bubra on imitative restenosis after percutaneous transluminal coronary angioplasty(PTCA) in cholesterolfed rabbits is studied systematcally on cell and molecule levels.Methods Establishing a model of restenosis by injuring carotid arteries in cholesterolfed rabbits. Observation of neointimal hyperplasia and collagen synthesis morphometrically, determining the extent of VSMC proliferation and macrophage in filtration by immunohistochemistry, measuring the level of blood lipid , surveying the level of Agll in plasm by radioimmunoassay and the response ofcarotid artery to Ag II by using a force transducer connected with a polygraph,detecting mRNA expression of NADPH Oxidase P22phox,MCP-l,MKP-l by in situ hybridization and transcription-polymerase chain reaction.Results Experimental results of our study are first submitted. On the imitative post PTCA restenosis in cholesterolfed rabbits.1 Radix Paeoniae Bubra(RPB) can modestly less neointima hype-rplasia and proliferation of collagen type I induced by balloon-injury, remarkable prevention the formation of restenosis and remodeling after balloon injuring carotid artery .2 RPB may increased acetylcholine(Ach)-induced vasorelaxtion. restorated the function of endothelium damaged by balloon injuring carotid artery .3 RPB can decreased the level of angiotensin D(Ag II )and aldosterone(ALD) in plasm ,weakened the response of carotid artery segments to Agll after balloon injuring carotid artery.4 RPB down-regulate expression lever of NADPH Oxidase p22 mRNA in vessel wall of injuring carotid artery significantly.5 RPB inhibits MCP-lmRNA expression remarkable, and decreased the degree of macrophages infiltration especially in vessel wall of injuring carotid artery.6 RPB up-regulate expression the level of MKP-lmRNA in vessel wall of injuring carotid artery markedly.Conclusions RPB intervention model successful established of restenosis by injuring vascular endothelium with balloon in cholesterol-diet rabbits is first submitted. Hypothesis of mechanism for RPB prevention the formation of restenosis after balloon injury is first submitted. Our study indicate that RPB can prevent the5formation of restenosis after balloon injury carotid artery by inhibiting the activating of renin-angiotensin system (RAS), NADPH Oxidase, monocyte recruitment and MCP-1 ,and blocked the MAPK signal pathway participating in proliferation of VSMC.and collagen type I . This action mechanism of RPB is studied first on RAS. oxidative stress Inflammatory reaction signal pathway aspect. Our studies provide useful scientific data of laboratory for clinic' application of RPB and mechanism for RPB prevention the formation of restenosis after balloon injury is first submitted.
