Purpose:To assess the effect of breviscapus (vant) hand-mazz erigeron (EBHM) on oxygen induced retinopathy (OIR) in a mouse model.Methods: Mouse models of oxygen induced retinopathy by 5 day exposure to 75% oxygen from postnatal day 7 to 12 were used to produce retinal neovascularization.On the one hand,EBHM(10mgand20mg/0.2ml/day)was intraperitoneadly administertered to mice pups of OIR from P7-P16 to determine if EBHM altered retinopathy . On the other hand. EBHM was administered from P17-P20 to determine if it cured retinopathy at the same condition.Fluorescein conjngated dextran angiography of retinal vasculature was employed and retinal whole mounts were repared to score features of the retinopathy.In addition quantification of retinal neovascular nuclei was caculated on the sections of HE staining.Results:Animals that were exposed to hyperoxia for five days had a median retinopathy score of 9.33±1.03 versus control animals that had a retinoathy score of 0.75±0.46 with P<0.01.Subscores for blood vessel growth, blood vessel tufts, extra-retinal neovasularization, central vasoconstriction, he-morrhage,and blood vessel totuosity were all significantly different between control and treated groups.In addition, quantification of neovascular nuclei showed a significant increase in the number of nuclei extending beyond the inner limiting membrane into the vitreous in hyperoxic treated animals(55.33±14.81 versvs 0.6±0.9P<0.01).EHBM at dose of 10mg and 20mg/0.2ml/day from P7-P16 and of 20mg /0.2ml/day from P17-P20 improved the retinopathy as measured by the total retinopathy score (6.16±0.75, 4.10±0.84, 6.20±1.04, respectively). Theaverage number, of extraretinal neovascular nuclei on six retinal sections was significantly decreased by EBHM at doses of lOmg and 20mg/0.2ml/day from P7-P16 and of 20mg/0.2ml/day from P17-P20 (37.67±10.59, 21.33±8.09, 36.00±10.15, respectinely)when compared to hyperoxic treated groups (55.33?4.80).Conclusion: EBHM maybe prevent and cure oxyen induced retinopathy in the mouse.The proliferation of retinal new vessel should be suppressed by EBHM.
